Ridgeway Science has a 30-year pedigree in producing gold-standard progesterone ELISAs, used by vets and within research institutes in the UK and overseas. Our ELISAs provide a fast and accurate measurement of an animal’s progesterone (P4) level in milk (milk progesterone), serum or plasma.
Ridgeway's ELISAs have also been used as a benchmark test to compare other new and upcoming on-farm aids for heat detection, such as our product P4Gold.
[Ridgewa] Progesterone ELISA test: Equine (Plasma/Serum)
ELISA test for measuring progesterone (P4) in equine serum or plasma utilises unique high-affinity anti-progesterone (P4) antibody which is highly specific. The kit consists of 96 well plate plus reagents and standards.
This test can be used for:
- Routine fertility management
- Checking for pregnancy loss
- Research into equine reproduction and its management
A small volume of serum or plasma is used as the test sample. It is recommended that each sample is run in duplicate and each standard run in triplicate. The substrate produces a colour change inversely proportional to the quantity of progesterone in the sample tested. This means a strong colour equals very low progesterone and vice versa. The colour change is read quantitatively with a plate reader.
Features of Ridgeway Science’s Progesterone ELISA test:
- A top-of-the-range monoclonal antibody with a high affinity for P4
- P4 range of approx. 0 -20ng/ml
- Low cross-reactivity with other steroids and hormones therefore no false results
- High throughput format with results in 3.5hrs or less (1 hr incubation recommended)
- Chromogenic substrate which is not light sensitive
- Long-established assay, cited in multiple equine research papers
- No dilution of samples necessary therefore can apply straight onto assay
- Small 10ul sample size
- Can be read at 405nm, 550 or 570nm with differing substrates (available at request). The best results are obtained at 550nm.
- Incubation times – 1.5 hours for the plasma assay.
Serum progesterone concentration in pregnant mares should stay above 6.3 nmol/L from day 18/20 until around day 200 of gestation.
No relationship has been proven between progesterone levels in pregnant mares and the integrity of pregnancy. Pregnancy is unlikely for mares with levels <2 ng/ml, typically pregnant mares have levels >4 ng/ml however this can vary greatly by day and individual. Despite not being an accurate pregnancy test, the progesterone assay can be a useful tool in monitoring mares with a previous history of repeated pregnancy failure.
Progesterone levels >2 ng/l (6.3 nmol/l) in non-pregnant mares indicate functional luteal tissue.
